HuffYUV.dll is just the codec file that allows you to save clips compressed as Huffyuv .avi's within editing programs or programs such as VirtualDub. Once that .dll is installed to its appropriate place, you shouldn't ever have to touch it again.

Besides the filters that come with the amvapp, you can get more filters from doom9 or this site

when you get new filters, you'll want to put them into C:\program files\avisynth2.5\plugins (it'l be easier for you if you do that)

and no, do not save it as huffyuv or huffyuv.dll
you save the script as [insert your script name here].avs

You must open the avs file in vdubmod then save as and change the codec at the bottom to huffyuv. Your file will be an editable avi file.

Some of the plugins will have documentation if you go to:
Start > All Programs > AMVapp > AviSynth Filters > Documentation

And there's even more in :
C:\Program Files\AviSynth 2.5\Docs\english\externalfilters
and
C:\Program Files\AviSynth 2.5\Docs\english\corefilters
... yea ...
