Life is Shattered...There is no need to go on...in this human-filthed planet....Escape is not possible. To try and wipe them out is absurd...they are like cockroaches....

Can't think of a video game one at the moment, but as for anime, (excluding DB/DBZ/DBGT), I'd have to say Brandon Heat/"Beyond the Grave" from Gungrave. Much as I hate the show, in the first episode he literally got pounded into the ground by some large bio-monster thing, and he just took it. Sucky character or no, he's still one tough guy.
Who knows what fires burn in the souls of others, or what burdens weigh upon their hearts?

By far the toughest anime character I have even seen is Gutts from Berserk. He is just bad ass.

On a tangent here, Raj Ahten, a villain from David Farland's 'Runelords' books is as close to invincible as I've ever seen a fictional character be. The world allows a magical transferrance of certain attributes to another person, called an 'endowment'. Raj Ahten uses charm, blackmail and bribery to accumulate said endowments, to the point were he now has most of the stamina, grace, voice, strength and glamour of the best part of the subcontinent from which he hails. He also has currently seven times the metabolism of human beings.
His shout can literally tear down castles, but he is so beautiful and persuasive that most just throw open their gates for him on demand. He has enough stamina that you can stab him through the heart and he won't even bleed- the wound heals as you remove the knife.
He's one tough customer.
