One of their videos is done in anime. So yes, Minami and the other guy are drawn as anime characters on stage and they're traveling across the countryside. I think it's for Reflection. The music video for Truth (theme for Detective Conan) is a CG animation, but you don't get to see Two Mix themselves in it. Definetely they acknowledge a large number of their fans are anime otaku.

Minami Takayama is a voice actress with a pretty good career. So she was part of Doco and Ribbon, for example. So you can see her if you get live action performances for the groups she was part of. And the music videos for Doco are of course for Ranma 1/2.

Try to listen to Ayumi Hamasaki, the best singer in Japan. Also try Boa, Hikaru Utada, V-6, T.M. Revolution, Hitomi, Aikawa Nanase, SES, See-Saw, Rikki, Judy and Mary, and many many more I can't think of rite top of my head.

The Pillows, Every Little Thing, Do As Infinity, Utada Hikaru, Ayumi Hamasaki, Boa, TM-Revolution, See Saw, Yoko Kanno, and there's more... I just have no idea what their names are ^^;

My favorite is the pillows ^_^
