I think that was the point. From what I've heard, in the novel it's based off, the author is careful to not use a gender-specific pronoun about kino for the entire time, and my guess is the episode that deals with her past is towards the end, so you don't no if its a he/she until then.I find that it didn't ever really matter what gender she was.

In the anime, they did use both pronouns for Kino. Still using the colleseum episodes, when she first arrived, they referred to her as "Mister Kino." Later on, she is referred to as her proper gender
