Elfen Lied was...hmmm...don't know how to classify it...not really a love story...but I did like that anime overall...I'd recommend it to the non-squiemish...
Here, I'm gonna be uber-specific for a second. I was trying not to appear to picky, but in the end just showed how picky I was......so, here's like exactly what I'm thinking of (anything close I'll take a look at):
- Non-comedy, non-ecchi
- Action good, but the action should be in the name of love! (...too corny?)
- More mature is good, although I'm not a big fan of gratituous gore or nudity. It should have meaning!
- I like: Angels, vampires, etc; I hate: mecha, cyborgs, incest, pedophilia, yaoi
- Tragedy. Guy falls in love with girl is boring. Guy loses girl, tries to get her back, fights for her, etc., whatever...everything doesn't have to have a happy ending either...

For something completely different, how about Gankutsuou? ("The Count of Monte Cristo", a-la anime).
Not a love story per se, but all of it is underpinned in a failed love (followed by, of course, revenge), and there are some great romantic sub-plots involved. And it hits big points on your seriousness criteria and, surprisingly, somehwat on vampires too.
And if that's not enough, the art is gorgeous.
Failing that - if you can get past many of its comedic features, KareKano is a very good, albeit incomplete, believable story of a boyfriend-girlfriend relationship. There's a lot of darker, non-comedic undertones in parts of that one. Also ranks highly in my book.

Gankutsou is one of my favorite anime ever...oh I'd love to find something on par with that...
I'm downloding Saikano and Hanbun no Tsuki ga Noboru Sora now, gonna take a look at those two first...
hmmm...is DN Angel a love story? It looks good but I just can't tell where it's at in tone and theme...

i dont alot of anime's with emotional love, but 3x3 eyes had its moments here and there, kinda like Inuyasha love, along with tons of blood.
another one was Tokyo Godfathers, im not 100% sure on it but Miyuki commented on it about Hana & Gin.

If you are interested in Voices of a Distant star, You might also try The Place Promised in our Early Days. Both are very good Voices is a story of love over time and space, Place is a romantic story of a boy trying to save his girl from a coma of uknown cause.
Kite: romance and violence.
Howl's Moving Castle: Laputa, Castle in the Sky: Whisper of the Heart: Princess Mononoke: The Cat Returns: all good romance stories from studio Ghibli.
I just got Mermaid Forrest, done by the creator of Inuyasha. it is a horror story series with a lot of romance. I haven't watched it yet so I can't say how good it is.

I'd recommend Kare Kano, a.k.a. His and Her Circumstances, if you hadn't ruled out comedy out of hand. It's everyday-slash-upbeat-slash-angst-with-power-plant, but "romantic" and "goofy" are the two biggest slices of the pie. Standard viewing for hetero romance that's actually romance and not "lol oops i fell on a boobie, repeat for 26 episodes."
To add on a previous comment: Piano doesn't have much romance. There's omg-secret-crush talk, but it's not focused on romance as a whole, and it's not tragic. Slightly angsty, but not tragic.
Your stats sound like either X or Angel Sanctuary without the incest to me, and hey, I'd like to find something along those lines myself. For me, Yami no Matsuei is in the same category, but that's shounen-ai. Haven't seen Tsukihime yet. (note: Tsukihime, not Tsukiyomi. Grownup chicks that are probably vampires, not moemoe lolis. Not my bag.)
